- 3:30 PM1hMCB Seminar Series: Joana PaulinoJoana Paulino Assistant Professor, Molecular Biology and Biophysics, UConn Health Host: Vikki RobinsonViral-Host Protein Interactions: Understanding Molecules to Undermine DiseaseVirus-host protein interactions are part of core events in the viral lifecycle. These interactions have been alluring targets for drug discovery due to their proposed reduced possibility of mutational events leading to drug resistance. However, the multifunctionality of viral proteins at different stages of the virus life cycle and the many functions each viral protein carries leads to many challenges in their biophysical and structural characterization. Examples of these challenging interactions are: (1) the HIV negative factor protein (NEF) interaction with human Serine Incorporator 5 (Serinc5) and, (2) the HIV viral protein U (VpU) interaction with the human cluster of differentiation factor 4 (CD4). These interactions are essential for HIV evasion of the cell's native immune response. More recently, our characterization of the interaction between the Zika virus (an Orthoflavivirus) non-structural protein NS4a and the human Ankle2 protein demonstrates how complex and many layeredare these interactions.About Dr. Paulino:Joana Paulino's research program combines biophysics, structural biology, and virology, studying fundamental principles of virus-host protein-protein interactions and physiological aspects of how these interactions lead to severe disease outcomes. Her past work focuses on characterizing viral and bacterial membrane proteins utilizing solid-state Nuclear Magnetic Resonance (ssNMR) and cryo-Electron Microscopy (cryo-EM). Currently, her laboratory focuses on how Orthoflaviviruses, such as Zika and Powassan virus, can overcome two of the most complex physiological barriers: the placenta and the blood-brain barrier. Paulino is interested in how Orthoflavivirus membrane proteins interface with the extracellular vesicle biogenesis pathway, the endoplasmic reticulum (ER) membrane, and cytoskeleton proteins, to alter cell membranes and the permeability of cell barriers, such as gap junctions.Paulino Lab website (https://health.uconn.edu/paulino-lab/)
