- Nov 2110:00 AMDoctoral Dissertation Oral Defense, Qingqi LinAbstract:Multiple Sclerosis (MS) is a chronic autoimmune disorder of the central nervous system, affecting an estimated 2.8 million individuals worldwide. Its underlying pathophysiology reflects complex interactions involving genetic susceptibility, immune dysregulation, and metabolic alterations. Emerging evidence has highlighted the potential role of the gut microbiome in MS; however, the temporal dynamics linking microbial communities with host immunity, metabolism, and dietary influences remain insufficiently characterized. To better understand these relationships, we carried out a six-month longitudinal multi-omics study including 49 participants: 24 untreated relapsing-remitting MS patients and 25 healthy controls matched by age, sex, and race. Our metagenomic analyses identified multiple gut microbial taxa significantly reduced in MS patients, as well as taxa showing positive and negative associations with clinical disability measures. These microbiome shifts coincided with marked alterations in peripheral immune cell composition.Although numerous studies have reported associations between gut microbial taxa and MS, the specific microbial signatures linked to the disease vary considerably across cohorts, leading to limited consensus. To address this inconsistency, we performed a meta-analysis of 16S rRNA gene sequencing datasets from seven studies that differed in geographic origin and technical protocols, totaling 524 adult participants (257 MS patients and 267 healthy controls). Each dataset was reprocessed using a standardized workflow to minimize methodological bias, and both study-level and pooled comparative analyses were performed. This integrated analysis revealed a set of microbial taxa that were consistently associated with MS across multiple cohorts, providing more robust evidence for reproducible gut microbiome alterations in MS.Several of the microbial taxa repeatedly detected across studies are noteworthy, either because of their consistent differential abundance between MS patients and healthy individuals or because of their potential metabolic interactions with the host. Elucidating the functional roles of these microbes will be essential for clarifying their contribution to MS pathophysiology. To support such downstream strain-level analyses—which require high-quality, contamination-free reads for reliable genome reconstruction—we developed StrainPolisher, a computational tool that identifies and removes contaminant reads from whole-genome sequencing (WGS) data at the strain resolution. Benchmark evaluations indicate that StrainPolisher delivers high accuracy and computational efficiency, providing a robust quality-control solution for microbial genomics workflows.Taken together, this work integrates longitudinal multi-omics profiling with cross-cohort meta-analysis to deepen our understanding of the gut microbiome's involvement in MS, and further introduces a generalizable strain-level quality control tool that supports robust microbial genomic investigations more broadly.

- Nov 2112:00 PM12 Step Recovery Meetings - Online12 Step Recovery Meetings - Two Offerings! 12 Step Recovery is a fellowship of people who come together to build an individualized program of recovery based on the program's 12 Steps of Recovery. This pathway includes a spiritual component, Higher Power, that is unique to each individual, and that may evolve or change completely over time. 12 Step Recovery at UConn is intended to provide exposure and practice with 1) 12 Steps overall with a strong focus on the first three Steps, 2) primary program components, literature, and guiding principles, and 3) building a community of support and fellowship. 12 Step Recovery is an abstinence-based program designed originally to support people living with addiction to substances; however, it has grown to include other types of addictions and problem behaviors.
